Mitochondria are essential subcellular organelles that regulate cell life and death. Best known as the “powerhouses” of the cell, they also play a central role at the intersection of innate immunity and programmed cell death pathways. Mitochondrial dysfunction has been linked to a wide array of both inherited and acquired human diseases. In this talk, I will discuss our approaches to characterizing and understanding primary mitochondrial dysfunction using cell and animal models of mitochondrial disease (MD)—a group of rare genetic disorders caused by mutations that impair mitochondrial protein function. Although our focus is on primary MD, the methods and workflows presented can also be adapted to study mitochondrial dysfunction in non-MD cellular and animal models.
